Covalys Biosciences AG, today announced the launch of new products for immobilization and pull-down experiments based on its proprietary SNAP-tag. SNAP-capture kits present a new way to
perform pull-down experiments by covalently immobilizing a protein of interest, without prior purification, on a resin or magnetic bead surface. The kits may also be used for studies of
protein function.
This novel approach dispenses with the potentially damaging capture of the protein on a chemically activated surface and the problem of drift associated with antibodies for protein
capture by offering a universally applicable covalent and orientated immobilization of proteins in physiological conditions. SNAP-capture Magnetic Beads Kits eliminate the need to carry
out repeated centrifugation steps thus facilitating processing of samples and minimizing sample loss. SNAP-capture Purification is intended for protein purification and permits
immobilization under mild conditions, followed by vigorous washing of the immobilized protein without fear of eluting the protein of interest as the interaction with the resin is
covalent. A new addition to the building blocks product range CBG-NH2 offers a further improved way to immobilize proteins to carboxy surfaces for applications such as Biacore.
SNAP-express Gateway plasmids offer a fast and easy way to insert the ORF of interest from a Gateway Entry Clone into a mammalian SNAP-express Gateway destination vector thus minimizing
or obviating the time required for cloning. SNAP-express Gateway plasmids are available for cloning the ORF to either the N or C terminus of the SNAP-tag and are supplied with a positive
control which directs a SNAP-tag fusion protein to the nucleus.
The introduction of the MCP-tag offers a novel and highly effective way to specifically label proteins on the surface of living cells. Once the protein of interest is expressed as a
MCP-tag fusion it can be rapidly covalently labeled by exposing the cells to a non-cell permeable fluorescent CoA Substrate in the presence of a specific labeling enzyme, SFP-Synthase.
This enables precise control of the timing of onset of fluorescence. In contrast to the autofluorescent proteins, the same fusion protein can be labeled with different fluorophores simply
by exposing the cells to different fluorescent substrates. The small MCP-tag can be used in combination with ACP-tag to visualize two different fusion proteins on the surface of living
cells by sequentially labeling proteins with CoA Substrates carrying different fluorophores and the appropriate Synthases (also available from Covalys). Labeling is highly specific and is
restricted to the cell surface therefore there is no background associated with internal pools of proteins. New CoA substrates for labeling ACP-tag and MCP-tag include CoA-Biotin for
selective biotinylation of cell surface proteins and CoA-SH, a building block containing a thiol group that enables customers to make their own CoA Substrates.

different fusion proteins can be visualized simultaneously on cell surfaces by virtue of these small protein tags."
